The cellular levels of O-glucosides of 3H-(diH)Z and 3H-(diH)[9R]Z, the major short-term metabolites of 3H-(diH)Z having been exogenously supplied to photoautotrophically growing suspension cell cultures of Chenopodium rubrum, decreased significantly during further culture, irrespective of whether the cells were maintained in the stationary phase or were transferred to conditions restoring cell divison. Metabolism of both compounds was more pronounced during the active growth phase than during the stationary phase. The O-glucosides were converted preferentially to polar compounds of as yet unknown nature, which were partly excreted into the medium. The cellular pools of both glycosides remained compartmented within the vacuole. In contrast to the O-glycosides, the small cellular pools of the aglycones 3H-(diH)Z and 3H-(diH)[9R]Z maintained their level during the experimental period of 30 days. Small amounts of the glucosides, as well as of the aglycones, were recovered from the medium and could have resulted from the lysis of a few cells. The results demonstrate, for the first time, that O-glucosides of cytokinins are not irreversibly deposited within the vacuole of plant cells but may serve to maintain a small, but more or less constant pool of extra-vacuolar, presumably cytosolic, aglycones. (DiH)Z and its derivatives could be demonstrated to be endogenous cytokinins of Chenopodium rubrum suspension cultured cells occurring along with those of the isopentenyladenine and zeatin types.  相似文献

The western white butterfly, Pontia occidentalis, has distinctly different wing phenotypes during spring and summer generations as a result of phenotypic plasticity (seasonal polyphenism). We experimentally generated different seasonal phenotypes in the lab by altering photoperiodic conditions during rearing, and released the resulting butterflies in the field. Mark-recapture studies were then used to estimate the effects of the polyphenism on activity patterns and adult survival in both late-spring (one study) and summer (two studies) conditions. There were no significant effects of rearing treatment on temporal patterns of behavioral activity during either the late-spring or the summer field studies. Recapture probabilities were consistently higher for males than females in all three field studies; in the summer 1992 study, recapture probabilities were higher for long-day (LD) than for short-day (SD) treatment groups. During the late-spring 1992 study, there were no significant differences between LD and SD treatment groups for survival probabilities. In the two summer studies, there were significant effects of photoperiodic treatment on survival probabilities; in the summer 1992 study, LD individuals consistently had higher survival probabilities than SD individuals; in the summer 1991 study, there was a significant interaction between treatment and time period on survival probabilities, such that survival probabilities were higher for LD than for SD individuals in two of four time periods. The consistent differences in survival probabilities between treatment groups in the summer 1992 study can be accounted for by the differences in wing traits between the treatment groups. Micrometeorological data from the study site showed that midday ambient temperatures averaged ~3°C hotter during the 1992 than the 1991 summer study and that ambient conditions during the late-spring 1992 study were relatively warm and sunny for the season. These results document the varying relationships between phenotype and fitness in the temporally fluctuating environments experienced by this population.  相似文献

With the goal of understanding how nervous systems produce activity and respond to the environment, neuroscientists turn to model systems that exhibit the activity of interest and are accessible and amenable to experimental methods. The stomatogastric nervous system (STNS) of the American lobster (Homarus americanus; also know was the Atlantic or Maine lobster) has been established as a model system for studying rhythm generating networks and neuromodulation of networks. The STNS consists of 3 anterior ganglia (2 commissural ganglia and an oesophageal ganglion), containing modulatory neurons that project centrally to the stomatogastric ganglion (STG). The STG contains approximately 30 neurons that comprise two central pattern generating networks, the pyloric and gastric networks that underlie feeding behaviors in crustaceans1,2. While it is possible to study this system in vivo3, the STNS continues to produce its rhythmic activity when isolated in vitro. Physical isolation of the STNS in a dish allows for easy access to the somata in the ganglia for intracellular electrophysiological recordings and to the nerves of the STNS for extracellular recordings. Isolating the STNS is a two-part process. The first part, dissecting the stomach from the animal, is described in an accompanying video article4. In this video article, fine dissection techniques are used to isolate the STNS from the stomach. This procedure results in a nervous system preparation that is available for electrophysiological recordings.  相似文献

Streamflow-related variability in nutrient flux represents an important source of uncertainty in managing nutrient inputs to coastal ecosystems. Quantification of flux variability is of particular interest to coastal resource managers in adopting effective nutrient-reduction goals and monitoring progress towards these goals. We used historical records of streamflow and water-quality measurements for 104 river monitoring stations in an analysis of variability in annual and seasonal flux of nitrate to the Atlantic coastal zone. We present two measures of temporal flux variability: the coefficient of variation (CV) and the exceedence probability (EP) of 1.5 times the median flux. The magnitude of flux variations spans a very wide range and depends importantly upon the season of year and the climatic and land-use characteristics of the tributary watersheds. Year-to-year variations (CV) in annual mean flux range over two orders of magnitude, from 3–200% of the long-term mean flux, although variations more typically range from 20–40% of the long-term mean. The annual probability of exceeding the long-term median flux by more than 50% (EP) is less than 0.10 in most rivers, but is between 0.10 and 0.35 in 40% of the rivers. Year-to-year variability in seasonal mean flux commonly exceeds that in annual flux by a factor of 1.5 to 4. In western Gulf of Mexico coastal rivers, the year-to-year variablity in the seasonal mean flux is larger than in other regions, and is of a similar magnitude in all seasons. By contrast, in Atlantic coastal rivers, the winter and spring seasons, which account for about 70% of the annual flux, display the smallest relative variability in seasonal mean flux. We quantify the elasticity of nutrient flux to hypothetical changes in Streamflow (i.e., the percent increase in flux per percentage increase in mean discharge) to allow the approximation of flux variability from streamflow records and the estimation of the effects of future climatically-induced changes in Streamflow on nutrient flux. Flux elasticities are less than unity (median = 0.93%) at most stations, but vary widely from 0.05% to 1.59%. Elasticities above unity occur most frequently in the largest rivers and in rivers draining the arid portions of the western Gulf of Mexico Basin. Historical flux variability and elasticity generally increase with the extent of arid conditions and the quantity of nonurban land use in the watershed. We extend the analysis of flux variability to examine several case studies of highly unusual meteorological events capable of significantly elevating nitrate flux and degrading estuarine ecology.  相似文献

Teachers' conscious priorities in selecting topics for teaching rarely include the ‘biology/social science interfaces’; neither are many usually aware of the existence and implications of ‘hidden ideologies’ in the curriculum. We typically observe large‐scale outcomes that arise from the interactions of many hidden, small‐scale processes. Examples include age of disease onset, rates of amino acid substitutions and composition of ecological communities. The macroscopic patterns in each problem often vary around a characteristic shape that can be generated by neutral processes. A neutral generative model assumes that each microscopic process follows unbiased or random stochastic fluctuations: random connections of network nodes; amino acid substitutions with no effect on fitness; species that arise or disappear from communities randomly. These neutral generative models often match common patterns of nature. In this paper, I present the theoretical background by which we can understand why these neutral generative models are so successful. I show where the classic patterns come from, such as the Poisson pattern, the normal or Gaussian pattern and many others. Each classic pattern was often discovered by a simple neutral generative model. The neutral patterns share a special characteristic: they describe the patterns of nature that follow from simple constraints on information. For example, any aggregation of processes that preserves information only about the mean and variance attracts to the Gaussian pattern; any aggregation that preserves information only about the mean attracts to the exponential pattern; any aggregation that preserves information only about the geometric mean attracts to the power law pattern. I present a simple and consistent informational framework of the common patterns of nature based on the method of maximum entropy. This framework shows that each neutral generative model is a special case that helps to discover a particular set of informational constraints; those informational constraints define a much wider domain of non‐neutral generative processes that attract to the same neutral pattern.  相似文献

To simulate the effects of forest tent caterpillar (FTC) defoliation on trembling aspen growth and mortality, an artificial defoliation experiment was performed over three years in young aspen stands of northwestern Quebec. Defoliation plots of 15 × 15 m were established on three sites, together with associated control stands of pure trembling aspen. In 2007, root collar diameters were measured and positions of all trees were mapped prior defoliation. Severe FTC defoliation was simulated for three successive years (2007–2009) by manually removing all leaves from all but 7–10% of the trees present in the defoliation plots. Yearly surveys of growth and mortality were conducted until 2010 to evaluate defoliation effects on defoliated as well as surrounding undefoliated trees. In absence of other factors, growth and mortality of trembling aspen decreased and increased, respectively, after defoliation. Our study further revealed that small diameter trees died after one year of artificial defoliation, while larger-diameter trees died after repeated defoliations. Distributions of tree mortality tended to be aggregated at small scales (<5 m), corroborating gap patterns observed in mature stands following FTC outbreaks. This experiment revealed that trembling aspen mortality can be directly attributed solely to defoliation. Repeated defoliations during FTC outbreaks have the potential to profoundly modify stand productivity and structure by reducing tree growth and increasing tree mortality in the absence of predisposing factors.  相似文献

区域生态安全格局构建对提升生态系统服务功能提供了重要路径,同时统筹各种生态要素进行生态保护与修复分区也是新时期做好生态修复的重要举措。以济南市为例,基于现状生态系统类型分布,聚焦生态本底和地质灾害敏感性的特征,基于形态学空间格局分析方法和自然保护区结合进行生态源地提取。采用夏季降水、植被覆盖度、坡度3个地质灾害敏感性因子修正基本生态阻力面。并采用最小成本路径方法(Least-Cost Path method,LCP)提取生态廊道,构建了市域的生态安全格局。采用电路理论进行生态关键区域(生态"夹点"和生态障碍点)的识别,进一步划分生态修复改善区,并对此提出针对性的生态保护修复策略和工程措施。研究表明:1)市域生态源地的个数为35个,面积为567.15 km2,主要类型为林地和草地。空间上主要分布南部山区。生态廊道818.42 km,平均廊道长度为12.99 km,廊道分布存在较为明显的空间分布差异性,整体呈现出"一屏、一带、三轴"的生态安全格局。2)识别的生态修复关键区包含生态"夹点"25处,历城区生态"夹点"分布最为密集。全市亟需修复的生态障碍点共34处,面积为6.90 km2,主要分布章丘区。生态改善区共识别2994.84 km2,近期亟需修复的面积为96.1 km2,主要分布在长清区、历城区、莱芜区。3)通过对比生态修复关键区和现状土地利用类型,因地适宜的制定了生态修复策略与工程措施布置指引方向。研究结果可为济南市国土空间生态修复规划提供一定的技术支撑,同时也可为其他地质灾害敏感性区域的生态修复规划提供指引。  相似文献
